Bioreactor for Tissue Engineering, Team Bioreactor Week 12 – April 12 to April 18, 2007 Team Members: Karen Chen – Team leader Richard Bamberg – Communications Rachel Mosher – BSAC Dustin Gardner – BWIG Client: Dr. Susan Thibeault Dept. of Otolaryngology, UW Medical School Phone: 608/263-6751, Email: [email protected] Advisor: Brenda Ogle Assistant Professor/ Dept. of Biomedical Engineering Phone: 608/265-8267, Email: [email protected] Problem Statement The aim of this project is to redesign and reproduce a pre-existing Bioreactor that is specific to the culturing of human vocal fold fibroblasts. The existing design models the vibration of a single vocal fold under tensile stress and provokes the elongation of fibroblasts and the subsequent formation of an extra-cellular matrix, characteristic of vocal fold fibroblasts. Our goal is to improve upon this model by simulating contact between two vibrating vocal folds and potentially finding a better substitute for Tecoflex, because its porosity is non-uniform and the pore size is too large. The bioreactor will be made from a T-flask with elastic vibrators immersed in a buffer, which will function as vocal folds. This device needs to be easily replaced with disposable parts, along with 2 pairs of artificial vocal folds and a capacity to generate (computer controlled) vibrations at a frequency (0-400Hz). Restatement of Team Goals The components should arrive during the week and through out next week, and we will be putting the parts together. The main focus will be working on the actuator and make it function, along with the other components attached. We also exchanged some ideas of the setup. We then went to the machine shop to obtain shop passes for the future work that will be done in the machine shop. z Most components have arrived. z Richard cut open and tested the brittleness of the T-flask. The flask managed to be cut open, and the rough sides could be filed to smoother surfaces to improve the texture of the sides. z Team talked with the advisor on Friday (Apr. 13) and talked about the individual components and their functions. z Richard has further worked on the flask and created the first prototype (not yet finalized). Statement of Team Goals This week’s goal is to work in the machine shop and properly cut open the flask. The components will be attached and fixed onto the cut open flask. The testing of the vibration frequencies will be done with a replacement of the Tecoflex, such as rubber band since the amount of Tecoflex is very little. The actuator should also be attached and tested for the whole bioreactor system. Difficulties The free samples are not arriving, thus we have to order the components without testing them with the samples. Also, it would take up more waiting time.
