Recitation 18 B cells We get antibodies from B cells Naive B cells bind to a speci c epitope of an antigen B cells with surface antibodies that can bind a speci c antigen selected for clonal expansion That is how we get so many so many antibodies This expansion is also known as activation How is the correct B cell progenitor naive B cells made VDJ recombination Creates shorter DNA sequences by choosing speci c V s D s and J s Stomatic hypermutation a high rate of mutation Sloppy joining when more or less DNA is added during VDJ recombination This is advantageous because we get more variability because it can cause frame shifts Using antibodies in the lab Used to visualize a speci c protein of interest The best way to see a protein is to stick an antibody onto it Steps 1 Inject a certain animal with the protein of interest 2 Bleed the animal or get the spleen in order to collect B cells 3 Fuse B cells with myoloma cells Myoloma cells are an immortalize cancer cell 4 This creates a hybridoma which is also immortal and makes antibodies 5 Screen all of the antibodies created by the different B cells in a multiwell plate for speci city against your antigen 6 Isolate the hybridoma that we want 7 Expand the hybridoma to produce lots of antibodies 8 Optional chemically conjugate a uorescent molecule onto the antibody
