Begin Purification of Environmental UnknownHypothesis:If a bacterium is stained, then it will be easier to identify it when using Brightfield illumintionMethods:Begin with preparing a dry mount of a culture of Bacillus sp. Use the methylene blue stain for this slide and observe it under 100X oil immersion without a cover slip. Next, prepare dry mounts of Gram negative rod and Gram positive rod. Use a Gram positive coccus to stain this slide and observe it under 100X oil immersion. Finally, put some Klebsiella pneumonia into one drop of congo red, but do not heat fix this slide. Stain the slide with Maneval’s stain and observe it under 100X oil immersion.Safety Concerns:1. The stains used on the slides will stain anything they touch 2. Wear gloves and safety goggles3. Do not get oil on the 10X and 40X power lenses4. Proper use of the microscope is necessaryRelevance:Learning how to stain slides will make the specimen easier to view under the
