Electrophoresis Gel Handout Study Sheet The purpose of this worksheet is to go into greater detail than was gone over in class Each part will be shown in a stepwise manner Page 1 Step 1 Name each of the alleles Each line separates the 4 alleles Let s call the 1st row A1 2nd A2 3rd A3 4th A4 Now that we have named the alleles we can get the genotypes Page 2 Going down the number line you will notice that some of the columns have either 1 dot or 2 dots Step 2 1 dot a homozygote 2 dots a heterozygote You can now name each genotype Column 1 A1 A4 Column 2 A2 A4 Column 3 A3 A4 Column 4 A4 A4 Column 5 A4 A4 Column 6 A4 A4 Column 7 A1 A4 Column 8 A4 A4 Column 9 A1 A4 Column 10 A4 A4 Column 11 A4 A4 Column 12 A4 A4 Column 13 A4 A4 Column 14 A1 A4 Column 15 A1 A1 Column 16 A1 A1 There are a total of 5 different genotypes Page 3 Step 3 Calculating the genotype frequencies Count the number of each genotype and put that number over the total number 16 of genotypes shown in the gel picture This will be the genotype frequency of each type You can check yourself by making sure all the fractions add up to 1 Step 4 Calculate the Allele Frequencies There are two ways to do this Since the creature is diploid there is two of each allele in each genotype meaning the total number of alleles is 32 1 You can count the number of each allele type 2 Or you can use the equation Example A1 Heterozygote containing A1 A1 A4 4 16 A2 A4 1 16 A3 A4 1 16 A4 A4 8 16 A1 A1 2 16 Here s option 1 Fr A1 8 32 Fr A2 1 32 Fr A3 1 32 Fr A4 22 32 Page 4 Here s option 2 Fr A1 2 16 2 16 8 32 A1 A1 A1 A4 Fr A2 1 32 A2 A4 Fr A3 1 32 A3 A4 Fr A4 8 16 1 32 1 32 2 16 22 32 A1 A1 A1 A4 A2 A4 A3 A4 You can also check yourself again by adding all the fractions up to make sure the sum is 1
