Discussion homework 2 You can use your notes, resources, and work collaboratively on these questions. You will submit your answers in the form of a “quiz” on Moodle. It is not really a quiz, it is just the easiest way for us to review your answers in Moodle. These questions will be graded for completeness (1 point per question) and we will provide feedback on your answers. So if you answer a question incorrectly, you’ll know through the feedback but points will not be deducted. For these questions, and especially for open response questions on exams, more writing is not necessarily better. Be concise and don’t provide explanations that aren’t asked for. Due by Sunday night – there are some questions here that require material from Wednesday’s lecture. Use this information for the next 3 questions This diagram shows the JAK-STAT cytokine signaling pathway. The JAK proteins are attached to the receptor kinase. When cytokine binds, the receptor dimerizes and the JAKS are phosphorylated. The JAKS then phosphorylate the STATs, which are transcription factors. 1. Would you expect cytokine to be a hydrophobic or hydrophilic molecule? Explain your answer in 1 sentence? 2. What would happen if ATP was depleted from the cell? Explain your answer in 1 sentence.Discussion homework 2 3. Explain (1 sentence) what would happen to the signaling pathway if a mutation in the receptor caused it to always be dimerized. Use this diagram for the following questions 4. You examine these cells in the lab and can measure the presence, location, and phosphorylation status (phosphorylated or not) of the above molecules., describe one thing could you measure in the cells to that would tell you that the signal is currently (or was very recently) bound to the receptor. Assume that you cannot simply “see” if the signal is bound to the receptor. There are multiple correct answers to this question, please just list one. 5. Describe one way that you could modify the above pathway to turn off the signal after gene expression occurs. S = Signal R = Receptor TK1= tyrosine kinase 1 PO4 = phosphate group TK2= tyrosine kinase 2 TFa = transcription factor a RNAp = RNA
