MICR Labs Week 3 TUESDAY Antimicrobial Susceptibility Test Exercise 7 3 PROCEDURE WORK IN PAIRS Each pair uses both organisms Follow the directions on page 532 of your lab manual You will only be performing the procedure with the MH plates and you will be spreading 100uL of undiluted culture instead of streaking diluted culture The following antibiotics will be in the refrigerator in petri dishes You will use sterile forceps to remove antibiotics disks and put them on your plate Each pair will use ONE of the provided Next lab you will need to compile class data for your assignment Note The organisms used in this experiment are Escherichia coli and Staphylococcus cohnii ABO Rh Agglutination Background Information Exercise 11 5 Experiment Supplement pages 18 19 PROCEDURE WORK INDEPENDENTLY Follow the directions on page 19 of the lab supplement Each table will test a different person and compile results The transfusion worksheet page 21 of the lab supplement is due at the end of the lab period and must be the original page from your lab supplement or it will not be accepted Safety Dispose of all waste in proper containers and clean spills with disinfectant Stay seated during experiment Be careful with incinerators alcohol and Bunsen burners Each class will use 20 MH plates 2 pair BLACK STRIPE 10 blood typing slides 2 table THURSDAY Gram Stain data sheet pgs 201 202 is due at the beginning of lab and must be handwritten on the pages from your lab manual or it will not be accepted Antimicrobial Susceptibility Worksheet supplement pg 23 Must complete during lab and is due at the end of the lab period or will not be accepted You must turn in the page from your lab supplement NO COPIES EXAM 1 Everyone must perform a streak plate you will have 50 minutes to complete the exam BIOCHEMICAL ACTIVITIES OF BACTERIA These tests will be used for identification of unknowns Triple Sugar Iron Agar Exercise 5 21 PROCEDURE WORK IN PAIRS Each pair will use all 3 organisms Follow the directions on page 403 of your lab manual Don t forget to stab AND streak You will not be using an uninoculated control tube Note The organisms used in this experiment are Escherichia coli Citrobacter freundii and Providencia rettgeri Lysine Iron Agar Exercise 5 22 PROCEDURE WORK IN PAIRS Each pair will use all three organisms Follow the directions on page 408 of your lab manual You will only be using three LIA slants and you will not have an uninoculated control tube Note The organisms used in this experiment are Enterobacter aerogenes Escherichia coli and Providencia rettgeri Safety Be careful with incinerators and dispose of materials properly The bacteria used in this experiment are known as potential enteric pathogens therefore take extra precaution Media Used per Section 30 TSIA per section 3 pair 30 LIA 3 pair Chemicals Used This Week Crystal Violet Health rating of 2 Fire rating of 1 Hazardous in case of eye contact irritant of ingestion of inhalation Slightly hazardous in case of skin contact irritant Safranin Health rating of 2 Fire rating of 2 Hazardous in case of skin contact irritant of eye contact irritant Slightly hazardous in case of skin contact permeator of ingestion Non corrosive for skin eyes or lungs Iodine Health rating of 3 Extremely hazardous in case of skin contact irritant of eye contact irritant of ingestion of inhalation Ethanol Flammability Rating of 3 Health Rating of 2 Reactivity level of 1 HIGHLY FLAMMABLE may cause eye or skin irritation can be fatal if swallowed Mueller Hinton Agar No known hazards Health fire and reactivity all have a rating of 0 Triple Sugar Iron Agar slants No know hazards Health fire and reactivity all have a rating of 0 Lysine Iron Agar slants No know hazards Health fire and reactivity all have a rating of 0
