MICR 3164 1st Edition Lecture 2• Difference between Defined and Complex Mediao For defined, you’ll know everything that is in it; it has minimal stuff. Complex media has one or more unknowns in it.• Difference between Selective and Differential Property o Selective allows some things to grow and inhibits others, while differential allows everything to grow but they look different. Also, the media changes color.• Difference between General Purpose and Enriched Mediao General purpose media is very open and allows most things to grow, while enriched media is general purpose WITH some pickiness added.• Definition of Pure Cultureo Only one culture in on the plate• Definition of Aseptic Cultureo Only what YOU want is on the plate. This is done by never spreading other bacteria• Definition of a Colonyo A single or a few of genetically identical organisms• Definition of Fastidiouso Needs extra nutrients to be able to grow. “Picky”• What are the Relative Sizes of Virus, Bacteria, Fungi, and Protozoa?o Viruses are measured in nm; bacteria in um; and fungi and protozoa are measure in mm.• Difference between mm, um, and nmo Nm = 10-9; um = 10-6; mm = 10-3• What is a Brightfield Light Microscopy?o Background is light, object you’re looking at is dark• What is a Darkfield Light Microscopy?o Light comes down and around. The background is dark and the object is light.• What is a Phase Contract Light Microscopy? These notes represent a detailed interpretation of the professor’s lecture. GradeBuddy is best used as a supplement to your own notes, not as a substitute.o Puts shadows on the microorganism to see more details• What is a Florescent Microscopy?o Stain it with a florescent dye. “UV Microscope”• What is an Electron Micrograph?o All we have to keep in mind is that it uses electrons• What is a Gram Stain?o Gram staining positive looks purple; Gram staining negative looks pink• What is a Spore Stain?o Spores show up green while the other cells come up pink• What is an Acid-Fast Stain?o Acid-Fasts are pink under a microscope and everything else looks blue• How do you calculate total magnification on a light microscope?o The ocular is 10x the objective• Difference between SEM and TEMo A scanning electron microscope looks at the surface of a cell while a transmission electron microscope can easily see what is in the cell, and we can see right through
