Biological Imaging Chamber Client: Lance Rodenkirch Team: Justin Lundell (Leader) Val Maharaj (Co-leader) Jeremy Schaefer (Comm.) Andrew Dias (BWIG) Michael Socie (BSAC) September 28 to October 4, 2007 Problem Statement In order to control the environment while using laser-based confocal and multiphoton imaging of live cells, an imaging chamber must be developed to be temperature and environmentally correct with perfusion options. Last Week’s Goals *After meeting Professor Ogle on Friday, the hope is to understand all of the design considerations. *It was this week’s goal to have decided on a final design to pursue after mid-semester. Everyone has a good idea of what the device should look like in their head. The goal for this week is to document our ideas so that we can do the decision matrix on Tuesday next week. Summary of Accomplishments *Had Team meeting with Professor Brenda Ogle *Had Team meeting with Professor Sean Palecek *Team had meeting Tuesday Oct. 2 to start design matrix This Week’s Goals *Think of more designs, especially for specific components. *Schedule another meeting with our client, Lance, to talk about more specifics *Think of more characteristics to add to design matrix Difficulties When we met with Professor Brenda Ogle, she expressed some concerns with our design components, or at least some things that she and Lance differ in opinion on. Professor Ogle would like another chamber built, instead of our team using last year’s design, which what Lance would like. We had the idea of making a new chamber, as well as using last year’s chamber, and somehow making our system components applicable to both designs. As of right now, it is just a thought. Professor Ogle also expressed concerns with our method of warming the carbon dioxide by bubbling it through water. While she admits it may work well, she wants us to make it safer for both human and machine. So, in short, come up with a better design than a Erlenmeyerflask on a hot plate. There were some other minor concerns, but nothing that should prove too difficult. Our meeting with Professor Sean Palecek proved to be insightful as well. One thing that we did not take into consideration was that pumping warm gas into a chamber on top of microscope may cause condensation on the microscope, which could be very harmful to the equipment.
